Paging through the rest of that book, I see a few others with "R" and others with other letters.

I did not find a legend at the front of the book.

Many locations would publish the list of marriage licenses issued daily or weekly in the local newspaper. I've often seen "Do Not Publish" for those that the parties asked that the news of their marriage not be made public. The "R" and other letters could be a code for some similar restriction.

This one, bottom left, from the same book has several letter codes, including "R." https://www.familysearch.org/ark:/61903/3:1:9392-S5WC-37?view=index&action=view&cc=1614804&lang=en

Thank you! I have seen those with Do not Publish. Do you know why they would have requested this?

"Do Not Publish" was sometimes used if a young couple did not wish their parents/family to know. Or, sometimes, it was used to keep the exact date of marriage a secret, if the bride was already expecting a child.
