John Kreis and his wife Barbara
I am looking for information on John Kreis and his wife, Barbara, who had a daughter by the name of Susanna. Susanna was born June 27, 1824, in Pennsylvania, and she died between 1870 and 1880 in Bartholomew County, Indiana. Would like to know who John’s parents were, and would like more information on Barbara. I think John Kreis might originally be from Germany.

Pennsylvania is a big place. Let's narrow it down a little more. I did a quick search and didn't find a suitable candidate with the information you provided. Meaning: I found a few Susanna's.
Is your Susanna in the FamilyTree? If yes, do you mind providing Susanna's ID number. Before I delve into figuring out how to best direct you, I want to be sure I have the right Susanna.
Since Susanna was born in 1824 (where do you have this date from?) her parents could be in the 1820 and should most definitely be in the 1830 census records. Does Susanna have siblings? Was she the first child, a middle child, or the last? if Susanna has older siblings, were they born in the US? Or in Germany? The census may only provide "Germany" or if you are lucky, a province. If you find John or Johann Kreis on the 1820 and 1830 censuses, you will have a little more information to work with.
I would then follow up with a census survey, look at any census records showing the Kreis family to see how the family grows and develops. With that information, you can now look at the 1820 census to see if there are any other families with the Kreis surname in the same area of where Susanna, and her parents lived. Using this FAN (Family, Associates, and Neighbors) approach may help you identify a family connection to another Kreis, who may have indicated where they are from. The early shipping records for this time period don't usually yield a lot of information on exactly where someone came from. I think one of your best bets, is to see if Susanna's siblings, or father, or mother ever had an obituary published in a local newspaper. Again, this is where knowing where Susanna's family lived becomes important.
In order to do the German research part, you will need to figure out which US records will help you answer your question about where your Kreis family came from. To learn more about Susanna's father, John (or Johann) you will need to research where Susanna was born and where her siblings were born to discover what those records reveal about the family.

I got her birth information from her baptismal record. She was baptized at Saint Paul Evangelical Lutheran Church in Garrett, Somerset County, Pennsylvania. That is where I found her parents. I have no clue if she has any siblings or not.
Her ID# is LCMG-JZC.

Just noticed on Susanna’s Find-A-Grave that her Dad’s name was Johann. I just changed it on Family Search and Ancestry.

It looks like Susanna was baptized as an adult. You could try poking around Somerset County, PA for more hints about any Kreis families living there. I would also look for a Pennsylvania genealogy group here on FS that could help you with the U.S. aspect of the research.
