This is from a Danish Parish register in 1853 for a marriage between Jacob Nielsen and Magdalena (Lene) Christensdatter. The question I have is regarding the word that precedes Lene in the attached image below.

I've seen this designation in front of other names but was unable to come up with the meaning of it.

Could it be as simple as Enke (widow)?

Yes, it is the definite form Enken ("the widow") of Enke.
