You've said '… I have 100s of letters (uploaded as PDFs containing pictures of the letters….' Are these already on FamilySearch somewhere? In your Gallery?

Good for you! Yes, though, this will take time and patience.
All I can really offer is the old adage: slow and steady.
Pull up each letter individually, zoom in, and start tagging each name.
I can't think of any more efficient way to do this than simply repeating this one process.
I hope others can share their top tips.

@BKBaird I'm not sure if it is any more efficient, but you could add one of your pdf's as a 'Memory' Source to one of the individuals highlighted in the pdf and in the process put also in your Source Box. Once it is there then you can add it to the various profiles also mentioned in the pdf. Using it as a Source will give you the added benefit of tagging the pdf if it contains, birth, death, residence information, marriages etc. You'd have to do the preliminary leg work to go thru each pdf to determine the PIDs of those mentioned. With the 'list' you can pretty quickly attach the pdf using 'Attach' in the Source Box.
If you're more ambitious you could get an automated transcription of each of the letters and use that to extract the 'key' bits and paste those in the 'Notes' field of the Source. Nice thing too is that if you make any kind of notes, it will be reflected for everyone the source is attached to….2 -
