In the meantime I have received a very detailed response from griffy and have incorporated his information into my original post - marked additions in green: as this group doesn't offer formatting in colour, I would ask anyone interested to view my post in the geneal-forum (no registration required) ... I will add just a summary in English below, incl. Hofmeister data and data from other sources.
First I would like to give you the links to the two important online sources griffy has used (and linked to):

Database of weddings in canton Zürich (16-18th century).
Now for the translation:
Vögeli - Erste Branche (a word I have never seen in a document in German - but likley means first branch).
Niclaus (1), the miller, lives before and after 1500.
Niclaus (2), the miller had a prebend in the "Spital" (for the more wealthy people more a residents' home than a hospital as we know it today). He married 1) 30 Mar 1529 Elisabeth Treyerin/Trayerin; 2) 29 Apr 1537 Barbara Gmünder.
Rudolf, the miller was elected "Zwölfer" in the guild Zum Weggen in 1535, in 1549 he was elected in the "Baptistalrat" (see below) as well as bailiff for Birmensdorf, in 1558 he became Kornmeister (responsible for supply and quality of grain) for the nunnery of Ötenbach. He married Verena Rütimann/Rüttimann and died 31 Dec 1565.
Heinrich, was born after 1500, in 1562 he was elected "Zwölfer" in the guild Zum Weggen (Hofmeister accidentally noted Guild of the millers, instead of the correct guild name), in 1567 he became bailiff of Andelfingen, in 1575 "Zeugherr" (either repsonsible for the city's military equipment or the quality of clothes in general), in 1586 bailiff responsible for the Augustiner monastry (which had been abolished in 1524 - the properties were converted to an "Amt" of city of Zürich). He died 13 Jan 1600. The notes on his marriages by Hofmeister are a bit confusing - proven by other sources are 1) 20 Dec 1568 Anna Heilerin; 2) 10 Jan 1582 Barbara Lindinner. Another marriage (prior to 1?) with Anna Studer is not documented by other sources.
Leonhard, was born after 1500, in 1571 he was elected "Zwölfer" in the guild Zum Weggen, in 1572 he became village mayor (where?), in 1591 councilman "mit Freier Wahl" (I guess, this implies not related to his guild position) and bailiff for Bülach, in 1594 bailiff for Ötenbach. He died 1596. On 12 Feb 1554 he had married Regula Hofmeister, daughter of Caspar Hofmeister and Barbara Risterin. Another marriage is mentioned without any detail.
Ludwig, was elected "Zwölfer" in the guild Zum Weggen in 1587, in 1598 he was elected in the "Baptistalrat" (see below). He died 08 Jul 1635 (likely outside Zürich, as his death was announced in Zürich 12 Jul 1635). He was married 1) 23 Jan 1581 Margareth Großmann; 2) 05 Oct 1601 Margaretha Brunnerin. See Stemmatologia Tigurina for further detail.
Regula, born 15 Mar 1581, marries 06 Sep 1596 (i.e. aged 15½) Rudolf Klingler, pewterer.

I only find a reasonably short description on how Zürich was governed in German on Wikipedia - let me translate the section on "Kleiner Rat" (the administrative council):
The "Kleine Rat" organised itself into two council groups, the "Natalrat" (Christmas Council) and the "Baptistalrat" (after John the Baptist). The 26 Natalrat members ruled from 25 December in the first half of the year, the 26 Baptistalrat members from 24 June in the second half of the year. Each council group had 13 Constaffel councillors - six nobles and seven burghers from the Constaffel - and 13 guildmasters.
The election of the 13 members of the Constaffel in each half-yearly council, who retained the hitherto customary designation of councillors (consules), was carried out by a commission of six members appointed by the mayor, two of whom had to belong to the nobility. These were opposed by an equal number of guild masters (scabini) in the "half-yearly council". The 13 craft guilds each elected a guild master for the two council groups, one governing and one standing still.

A question: you say 'had a prebend in the "Spital" '. It's a little unclear to me what you mean by "prebend." Does that mean that he was temporarily living there?
